    Research Topic

      유산균 은 “GRAS” (Generally Recognized As Safe) 로써 오랜 역사를 두고 발효 유제품을 중심으로 각종 발효식품, 장 류, 주류, 김치, 의약품 및 가축의 사료첨가제 등에 이르기까지 인류생활에 광범위하게 활용되어 오고 있다.


      식품미생물학 실험실은 식품 내 존재하는 미생물에 대한 연구와 이들을 제어하기 위한 연구를 실행하고 있다. 특히 Lactic Acid Bacteria (LAB)가 생성하는 생리활성 물질에 관한 연구에 중점을 두어 실시하고 있으며 이를 식품산업에 접목할 수 있도록 우유 및 유제품에 관련된 연구도 연계하여 진행하고 있다.




      유산균 유래의 생리활성물질은 그 역가가 치료 용의 의약품보다 낮다 하더라도 항상 섭취가 가능하다는 장점이 있다. 또한 화학적 합성물질이 아닌 천연 물질이므로 안전성과 함께 가공식품 내 신속히 적용 가능하다는 점에서 이와 같은 연구는 식품산업을 고부가가치 산업으로 발전시킬 수 있을 것으로 생각되며, 기능성 신제품의 개발 촉진 및 바이오 식품산업의 제품개발 활성화에 기여할 수 있을 것으로 기대한다.




      본 연구실은 mammalian cell culture를 위한 실험실 (식품미생물학 실험실 III, L210호), DNA manipulation 및 각종 Bacteria Culture 등 일반실험을 위한 실험실 (식품미생물학 실험실 I, 223호), 총 2개의 실험실을 분리하여 운영하고 있다. 현재 석/박사 학생들이 열심히 연구에 전념하고 있다 ( 식품미생물학 실험실 II, 214호, 3290-3491 ).

    1) 다양한 근원에서 분리한 유산균의 probiotics 특성 연구




      본 연구실은 다양한 근원에서 분리한 유산균주 및 병원성 미생물 균주를 보유하고 있으며, 우유 및 유산균이 생성하는 다양한 생리활성 물질의 정제, 특성 연구 및 대량 생산, 유산균 유래 물질에 의한 여러 가지 기능성을 지닌 신개념 바이오 식품 소재 탐색 관련 연구를 진행하고 있다.

    2) 유산균의 probiotics 로써의 기능성 연구


    최근에 유산균의 probiotics 로써 기능성을 입증하기 위한 in vitro 및 in vivo 실험을 다양하게 실시하고 있다.




    - 면역조절 작용

    Probiotics 균주에서 분획한 물질을 이용하여 염증성 장 질환 (Inflammatory Bowel Disease, IBD) 및 과민성 대장증후군 (Irritable Bowel Syndrome, IBS)과 관련된 면역조절 작용에 대한 연구를 진행하였다. 장 상피세포 모델인 대장 암 세포주 (HT-29 cell line)를 대상으로 세포 수준에서의 면역 조절 효능을 검증했으며, 세포주의 단백질 발현 패턴을 분석, 또한 동물모델 (DSS model)을 통해 숙주의 면역세포 및 anti-inflammatory cytokine의 분비를 살펴보았다. 그리고 유산균이 함유된 드링크 요구르트의 섭취를 통한 과민성대장증상 환자의 다양한 증상완화 평가를 위한 임상실험을 통해 장내균총 변화 및 cytokine, enzyme activity 등을 살펴보았다.




    - 골다공증 예방

    골다공증은 폐경 후 발생하는 대표적인 Bone Disease로, 조골세포 (osteoblast)와 파골세포 (osteoclast)의 osteocyte 불균형 및 칼슘 섭취, 흡수의 저하로 인해 발생한다. 골다공증 치료제로써 호르몬제 (estrogen), Bisphosphonate 등이 사용되고 있지만, 장기간 복용 시 많은 부작용을 가지는 단점을 가지고 있다. 이러한 단점을 극복하기 위해, Probiotics 균주가 발현하는 단백질 및 유청단백질 (WPC)을 이용하여 osteoblast cell의 분화 및 증식을 향상시키는 작용에 대한 연구를 진행하였고, 골다공증 동물모델(난소적출, OVX model) 을 이용하여 골밀도 및 골강도 향상, micro-CT, 혈청 내 ALT, AST, Creatinine, calcium, BUN, ALP, ACP 등의 변화를 통해 Probiotics 및 WPC의 골다공증 예방 및 치료 효과를 살펴보았다.




    - Biofilm 관련 연구

    대부분의 미생물이 자연상태, 식품가공공정, 인체 내에서 biofilm 을 형성하고 문제를 발생하는데 이들의 기작을 알아내고 문제를 예방하기 위해 biofilm 생성 시 발생하는 변화를 proteomics 등을 이용하여 연구하였다. Escherichia coli O157:H7 이나 Listeria monocytogenes 의 경우 빈번한 biofilm 의 형성으로 식품가공공정이나 식품의 표면에서 문제를 야기하는데 이를 연구하기 위해 biofilm 이 표면에 생성되는 기작과 probiotics 균주 처리 시 효능에 대해 연구하였다.




    - Cholesterol 저감화

    높은 cholesterol 수치는 동맥경화의 주요원인으로 현대인의 건강을 위협하는 가장 큰 문제 중 하나이다. 본 실험실에서는 김치와 같은 발효식품에서 분리한 유산균을 이용하여 in vitro 및 in vivo 실험을 통해 cholesterol 수치를 낮추는 Probiotics (생균제)를 개발하고 있다.




    - 천연 항균제 (Bacteriocin) 관련 연구

    항균작용이 뛰어난 Bacteriocin을 생성하는 유산균주 선발을 통해 천연 항균제 개발을 연구 중에 있다. Staphylococcus aureus, Streptococcus aureus, Listeria monocytogenes, Yersinia, Escherichia coli, Salmonella 등 식품 유래 병원성 미생물에 대한 항균작용의 기작 규명에 대한 연구를 진행하고 있다. 또한 Bacteriocin의 상업적인 생산 수율과 관련하여, Bacteriocin Production과 관련된 외부 인자에 대한 영향 및 Bacteriocin expression, regulation과 관련된 gene profile 연구를 진행하고 있다.
