    ▶ 세포외 공간에서 존재하는 나노튜브 : 본 연구실은 아래 그림과 같이 생쥐 일차 간세포 (mouse primary hepatocytes) 의 세포외 공간에서 세포와 세포를 연결시키는 수많은 나노튜브관을 위상차 현미경으로 최초로 관찰하였음 . 이 관은 액틴 필라멘트와 지질래프트막으로 구성되어 있기 때문에 터널링 나노튜브 (Tunneling Nanotubes,TNT) 라고 함

    ▶ 새로운 연구를 할 것인가 ? : 많은 연구자들은 우연히 TNT 를 관찰하여 왔으나 무시하고 지나쳐 왔음 . TNT 논문을 보고 나서야 “내 실험실도 TNT 를 관찰했었는데 ... ”하면서 후회함 (Bryant, Curr. Biol., 17, R655, 1999). 빈약한 정보와 모든 것을 새롭게 시작하여야 하는 두려움 때문에 많은 연구자들은 더 이상 연구를 진행하지 못하였음 . 

    ▶ TNT 연구의 네 가지 핵심 질문들 : 포유동물세포에서 TNT 가 처음 발견된 지 12 년이 지났지만 다음과 같은 네 가지 핵심 질 문들은 해결되지 않고 있음 . 본 실험실은 이 질문에 도전하고 있음 . 
     • TNT 는 어떤 기전으로 형성되는가 ? 
     • TNT 의 구성성분은 무엇인가 ? 
     • TNT 의 생리학적 기능은 무엇인가 ? 
     • TNT 의 의생명학적 응용은 가능한가 ?

    Research Area

     원형질막에 호르몬 수용체와 신호전달물질들이 제멋대로 흩뜨려져 있다면 세포신호전달은 느리고 비효율적으로 일어날 것이다 .

    따라서 원형질막에는 신호전달물질들을 조직적으로 모아서 세포신호전달의 중심지로 작용하는 지질래프트 (lipid rafts) 가 존재한다 .

    본 연구실은 지질래프트의 비교 단백질체학으로 당뇨병 , 근육병 , 노화 , 암 등의 원인이 될 수 있는 신규 신호전달단백질들을 동정하였다 .

    이들 지질래프트 단백질들의 기능을 이해하기 위해서 아데노바이러스 , siRNA, 유전자 적중생쥐 등 최신의 분자세포생물학적 실험기법을 사용하고 있다 . 특히 근육분화 동안 발현되기 시작하는 TRIM72/MG53 가 IRS-1 의 유비퀴티네이션을 유도하여 근육분화를 음성적으로 조절하기 때문에 TRIM72/MG53 적중 생쥐에서 인슐린저항성이 사라짐을 증명하였다 .

    더불어 암세포 , 노화세포 , 지방세포 , 근육세포에서 특이적으로 발현되는 지질래프트 단백질들을 다수 동정하여 이들 단백질들의 기능을 연구하고 있다 .

    본 연구실은 지질래프트 연구를 수행하는 중 지질래프트 단백질인 CD133 이 터널링나노튜브 (TNT) 의 형성에 중요한 역할을 수행한다는 것을 증명하였다 . 현재 CD133 에 의해서 만들어진 TNT 의 생물학적 기능연구에 모든 인적 - 물적 자원을 쏟아 붓고 있다 . 
     최근 사이언스 학술지는 지질래프트 분야를 미래의 노벨상 분야로 선정하였다 (Mysteries of the Cell. Science, 334, 1046-1051, 2011).
