    The Director of the research center of Tunneling Nano Tubes 

     

     

    Discovery of Extracellular Nanotubes

    ▶ For the first time, We discoverd innumerable fine tubes which connect cells each other in extracellular space of mouse primary hepatocyte ex vivo by confocal microscope. These tubes are known 'Tunneling Nano Tubes(TNT)' and they basically consist of actin filament and lipid raft.

     

     

    4 key questions for TNT research

    ▶ Although It is about a decade after TNTs were found in mammalian cells, those 4 core questions have not been solved yet. We have been challenging them. 

     

    • • How TNTs are conformed?
    • • What proteins compose TNTs?
    • • What is the function of TNTs?
    • • Is that possible to use TNTs for medical application?
    •

    ▶ For solving thoes questions, We organized the research center of Tunneling Nano Tubes with other 10 professors in division of Life Sciences, Korea University.

    1. Adult Stem Cell of Liver 

     Liver Regeneration is one of the reseach areas which mechanisms have not been elucidated. There are two major types of liver cells. Hepatocyte, the parencymal cell of liver and cholangiocyte composing bile duct. It has been argued that which type of cell is responsible for regeneration in liver injury because a lot of researchers have showed different results. We have been researching the type of cell and the role of proteins in liver regeneration carring out partial hepatectomy and drug treatment like CCl4, DDC and MCDE to genetically modified mouse.  

     

    2. Lipid raft and Tunneling Nano Tubes

     If receptors and signaling molecules are randomly scattered in plasma membrane a lot of signaling pathways are slow and  inefficient. Hence, there are lipid rafts to organize those signal molecules and are responsible for the center of signaling. We have identified a few novel proteins in lipid rafts related to diabetes, senescence, cancer and muscle disease. For these research, we used a bunch of  biological tools like adenovirus, siRNA and also genetically modified mouse. Especially, we figured out that insulin resistance was removed in musle of TRIM72/MG53 knock out mouse because the TRIM72/MG53 negatively regulated muscle differenciation inducing IRS-1 ubiquitination. 

     Recently, we found CD133, one of the proteins in ripid rafts, are important to conform the Tunneling Nano Tubes(TNTs) and have been researching the biological role of it.

     

    3. Function of Prominin1 in Liver

     Prominin-1 (Prom1) is a major cell surface marker of cancer stem cells, but its physiological functions in the liver have not been elucidated. We analyzed the levels of mRNA transcripts in serum-starved primary Prom1+/+ and Prom1-/- mouse hepatocytes using RNA-sequencing (RNA-seq) data, and found that CREB target genes were down-regulated. This initial observation led us to determine that the Prom1 deficiency inhibited cAMP response element binding protein (CREB) activation and gluconeogenesis, but not cyclic AMP (cAMP) accumulation, in glucagon-, epinephrine-, or forskolin-treated liver tissues and primary hepatocytes, and mitigated glucagon-induced hyperglycemia. Because Prom1 interacted with radixin, the Prom1 deficiency prevented radixin from localizing to the plasma membrane. Moreover, systemic adenoviral knockdown of radixin inhibited CREB activation and gluconeogenesis in glucagon-treated liver tissues and primary hepatocytes, and mitigated glucagon-elicited hyperglycemia. Based on these results, we conclude that Prom1 regulates hepatic PKA signaling via radixin functioning as an A kinase-anchored protein (AKAP).
