    암줄기세포의 발생과 특성을 조절하는 신호 전달 기전 연구

     

     

    종양 조직 내 복잡한 군집을 형성하고 있는 세포들 중 일부 줄기세포의 특성을 가진 미분화 암세포만이 종양을 형성할 수 있다는 사실이 알려지게 되었습니다. 암줄기세포라는 이름으로 불리는 이들 세포들은 주어진 환경에 종양조직의 대부분을 차지하는 세포들로 분화되어 종양조직 자체를 형성 할 뿐 아니라 항암제나 방사선 치료에 저항성을 가지고 있어 종양재발의 원인이 되는 세포로 알려져 있습니다. 이러한 암줄기세포의 근원이 되는 세포의 발생 원인을 밝히는 연구로서 성체줄기세포나 전구세포와 같이 지속적으로 분열 가능한 세포의 특정 유전자에 돌연변이가 발생하면 암줄기세포로 발전되는 것이 가능하다는 것과 함께 분화된 암세포에서도 종양유전자의 활성과 암억제유전자의 기능 손실에 추가적으로 줄기세포 특성 결정인자의 에피제네틱 발현 변화가 유도됨에 따라 암줄기세포로 역분화 또는 리프로그래밍이 될 수 있는 현상에 대한 연구를 진행하고 있습니다.


     

    암줄기세포와 표적 맞춤형 치료법 개발 연구

     

    기존의 암 치료방법은 종양 조직의 성장 억제 및 사멸을 목표로 한 방법으로 이러한 접근법은 종양 조직내의 대다수를 차지하고 있는 분화되어 종양 형성능이 없는 세포들에 선택적으로 작용하지만, 새로이 종양을 형성할 수 있는 암줄기세포에는 효과가 한정적이라는 문제가 있었습니다. 또한 이로 인해 기존의 종양치료가 악성 암줄기세포 군집의 성장을 유도하여 종양 재발을 촉진할 수도 있다는 가능성 마저 제시되고 있습니다. 따라서 본 연구실에서는 암줄기세포를 특이적으로 사멸시키는 것이 종양 재발과 타 조직으로의 전이를 억제할 수 있을 것으로 생각하고 암줄기세포와 이들의 발생 및 특성 조절에 중요한 표적 분자를 탐색하고 이를 암줄기세포 표적 약물전달시스템에 응용하고자 하는 연구를 수행하고 있습니다.
