    1. 세포노화의 기전과 암치료에 대한 응용

    세포노화는 세포자멸사와 함께 암발생에 대한 세포의 주요 방어기전이다. 예를 들면 Ras oncogene이 활성화되었을 때, 정상 세포는 암화되기 보다는 세포노화가 일어나 증식을 멈추게 됨으로써 암발생이 방지되며, 세포노화를 극복하였을 때 비로서 세포의 증식과 암화가 진행되게 된다. 그러나 최근에는 암 발생 후 치료 과정에서 화학요법제에 노출된 암세포가 대부분 사멸하게 되지만 일부는 스트레스성 세포노화를 겪고 살아남게 되며 이러한 세포가 암 줄기세포의 성질을 획득함으로써 세포노화가 암의 재발, 항암제 내성, 전이등에 기여하는 것으로 의심받고 있다. 그러므로 세포노화 기전의 이해는 암 발생기전의 이해는 물론 성공적인 암치료 전략의 개발을 위해서도 필수적이라고 할 수 있다.

     현재 우리 실험실에서는 암세포의 세포노화기전을 연구하고 있으며, 여기서 p38 MAPK, NADPH oxidase 활성화에 의한 산화스트레스와 함께 소포체 스트레스(ER stress)가 중요한 역할을 하는 것을 밝혔고, 소포체 스트레스에 의한 unfolded protein response를 매개하는 3가지 센서 분자중 ATF6α가 세포노화를 매개하는 역할을 하는 분자임을 규명하였다.


    2. 인삼 panaxydol 성분의 항암 작용과 기전

    인삼의 C17 polyacetylene 성분들은 항암효과를 가지는 것으로 알려져 있다. 우리 실험실에서는 이들 중 panaxydol이 암세포의 세포사멸을 유도하는 분자 기전을 규명하였다. panaxydol은 EGFR 활성화와 소포체 칼슘 방출에 의한 세포내 칼슘농도 증가, JNK와 p38 MAPK 활성화 및 NADPH oxidase 활성화, 소포체 스트레스에 의한 PERK 활성화를 통하여 미토콘드리아 경로의 세포자멸사가 유도됨을 보고하였다. 또한 panaxydol은 생쥐 암모델을 이용한 실험에서도 뛰어난 항암효과를 항암치료제로 개발될 수 있는 가능성이 높다.
